Offer Description

3-year PhD project for doctoral student in the H2020 Doctoral Network “LIPIDs as promising BRidge between novel disease InsiGHts and improved Tackling of disease” (LipidBRIGHT) in Montpellier (France).

Recent insights, from the LipidBRIGHT consortium, revealed preventive, diagnostic and therapeutic opportunities for lipids in CVD, chronic inflammation, chronic kidney disease (CKD) and cancer. Pro-resolving lipids in particular have been shown to reduce vascular calcification, a complication of CKD. In RD-Néphrologie, we have previously validated anticalcifying and antifibrotic properties of novel drug candidates in preclinical studies and aim to apply our pipeline to promising lipid candidates.

Through unique interdisciplinary cooperation and synergistically combining insights and technologies, 11 doctoral candidates in LipidBRIGHT will advance insights into lipid contribution to disease processes, improve lipid detection technology and study the impact of lipid therapeutic targeting and application, in a network of over 20 European institutes. Your objectives are

• To investigate the potential of pro-resolving lipids in preventing vascular calcification in the context of CKD, CVD and chronic inflammation in animal models,
• To decipher the underlying mechanisms of action using animal, ex vivo and in vitro models

What you will do

• You will develop in vitro models (hAoSMCs culture) and ex vivo models (aortic ring culture) of vascular calcificationto test pro-resolving lipid activities.
• You will design and perform preclinical studies with CKD and/or vascular calcification rat models receiving pro-resolving lipids.
• You will assess the protective capacity of lipids on vessels (vascular calcification, inflammation, fibrosis, osteoblastic differentiation), kidney and heart (function, fibrosis, inflammation), as well as inflammatory cells (inflammatory mediators, macrophage infiltration/subset changes and neutrophil phenotyping).
• You will also evaluate the effects of protective lipids on inflammatory processes in established ex vivo and in vitro assays, and the on neutrophil NET production and resulting fibrin and clot formation.
• You are employed by RD-Nephrologie research company, and supervised by Prof Angel Argilés and Dr Nathalie Gayrard
• You are hosted at BC2M academic laboratoryof University of Montpellier, and co-supervisedby Dr Karima Mezghenna

Working and training with LipidBRIGHT

You will attend yearly meetings organised by LipidBRIGHT, as well as international and national conferences.
