Overview

We are seeking a redeployed postdoctoral researcher to develop a novel, inexpensive method for extracting Mycobacterium tuberculosis DNA from clinical samples without mechanical equipment. The project will involve CL2/CL3 pathogen handling, molecular cloning, and bacteriophage manipulation.

Main Activities
  • Develop a lysis solution using pore‑forming short cationic peptides and a mycobacteriophage that disrupts both host and Mycobacterium cell membranes, enabling DNA release into a buffer compatible with downstream NAAT processing.
  • Assess the robustness and sensitivity of the lysis solution across sample types (tongue swabs, exhaled breath condensate, sputum) using quantitative microbiological methods.
  • Maximise sensitivity through enrichment of Mtb bacilli by adding a mycobacteriophage capture step before lysis and exploring various lytic agents.
