Job summary

Axle in Rockville, MD seeks a Postdoctoral Fellow to establish protein-focused capabilities, linking organoid phenotypes to molecular mechanisms. Implement APB-Display for rapid production and screening of protein libraries, and SMA/SMALP workflows for detergent-free extraction of membrane proteins into native nanodiscs.

You will work with imaging, spectroscopy, toxicology, and core facilities, document SOPs, and prepare for an independent research career in academia, government, or industry.

Qualifications

• Doctoral degree in biochemistry, molecular biology, chemical biology, or a closely related field.
• Hands-on recombinant protein expression and purification experience; cloning and binding assays helpful.
• Strong understanding of protein structure-function, membranes, and experimental design.

Responsibilities

• Biochemical support for organoid projects: expression, purification, conjugation, and assays.
• APB-Display: dual-functional hydrogel beads, DNA libraries, emulsification, PCR, transcription/translation, SNAP capture, fluorescence readouts.
• Library design for nanobodies and affinity reagents informed by targets and prior data.
• Quantitative screening: APB‑SortSeq and APB‑TiteSeq with controls for binding and expression.
• Orthogonal validation using FP, ELISA, pull-down, BLI, SPR, SEC, and other assays.
• Membrane-polymer workflows: SMA/SMALP and related polymers in cell lines and organoid models; optimize for native lipids.
• Nanodisc QC: fluorescence, dynamic light scattering, SEC, EM prep, MS-compatible workups.
• Membrane-complex enrichment: extract and enrich membrane proteins from membranes; preserve for downstream analyses.
• Nanobody workflows: labeling, concentration, purification, perturbation for imaging and analysis.
• Documentation and cross-training: SOPs, inventories, criteria, sample metadata, and training across the team.
