Parkinson’s disease (PD) is the second most common neurodegenerative disease. Most PD cases PD are considered idiopathic. However, more than 20 genes have recently been identified that cause monogenic forms of PD. PD was originally thought to predominantly affect dopaminergic neurons, but it is increasingly clear that PD-related mutations cause significant changes in inflammatory cells. This observation aligns with recent genetic studies indicating that, in PD, like Alzheimer’s disease, immune dysfunction is causally involved.
