We are seeking an experienced Bioanalytical Scientist for our Cambridge site with strong expertise in Mass Spectrometry and bioanalytical assay development to establish VidaVinci’s assay and analytical platforms. This individual should be highly motivated, creative and ideally have expertise in mass-spectrometry based analysis of both small molecules and peptides. Experience with Orbitrap Thermo-Fisher mass spectrometers instruments is a plus. The candidate should also have extensive knowledge of analytical software for the processing and interpretation of mass spectrometry data; familiarity with programming languages and database software is a plus.
Responsibilities
- Develop and implement state-of-the-art mass spectrometry-based analytics techniques to identify and characterize proteins, peptides, and protein ligand complexes.
- Develop, or oversee the outsourced development of, customized software for mass spectrometric analyses tailored to the specific demands of VidaVinci.
- Maintain all mass spectrometry instrumentation and the associated analytical software.
- Design, optimize, and execute technical development experiments to push highly multiplexed, isobaric resolving, de-novo peptide sequencing capabilities.
- Design, optimize, and execute affinity selection assays to characterize peptide interactions with target and off-target proteins (affinity purification, ALIS, CETSA/TPP) to support SAR investigations.
- Design, optimize, and implement additional multiplexed drug characterization assays. Utilization of qualitative methods where appropriate for differentiation of isomeric peptide, coupled with semi-quantitative methods to deliver on characterization needs.
- Collaborate with data teams to enable the generation, manipulation, and storage of data for informing future library designs.
- Work with collaborators, both internal and external, to effectively generate data and lead in assay development and execution to progress multiple targeted programs.
- Communicate results to the company’s senior leadership.
- Analyze experimental data and present it to a multifunctional team.
- Educate non-experts on the limitations of LCMS techniques and chemical incompatibilities to maintain instrument health and realistic analytical expectations.
- Proactively evaluate new technologies, be versed with current literature, and implement new methods into the mass spectrometry assay/analytical pipeline as necessary.
Required Qualifications
- Ph.D. in Analytical Chemistry, Chemistry, Biochemistry, Chemical Biology, or a related field with 5+ years of industry experience; OR M.S. with 6+ years of relevant analytical experience; OR B.S. with 6+ years of relevant analytical experience.
- Proficiency in mass spectrometry operations and theory required.
- Hands-on experience with bio-analytical chemistry assay development with mass spectrometry as a readout.
- Experience in sample cleanup strategies employed in LCMS workflows.
- In depth ability to troubleshoot LCMS systems and advise cross-functional teams about input compatibility.
- Knowledge and appreciation of laboratory safety procedures.
- Ability to thrive in a fast-paced dynamic team environment and be inspired to develop and apply new technologies to advance projects efficiently.
- Excellent written, organizational, analysis, and documentation skills.
- Ability to communicate progress effectively with colleagues across several scientific disciplines.
- Good working knowledge of adjacent domain(s) of drug discovery.
Preferred Qualifications
- Experience with Thermo MS software, their line of Tribrid mass spectrometers, and their routine maintenance. Ability to navigate MS method architectures, weigh the pros and cons of different scan setups, and enable sufficient data quality while enabling high sample coverage.
- Experience handling large-scale quantitative or semi-quantitative analytical chemistry assays.
- Experience scaling laboratory operations to enable greater sample throughput and experience scaling (or collaborating with people who scale) data analysis techniques to match demand.
- Hands-on experience with classical proteomics protein/peptide characterization workflows (peptide mapping, de-novo peptide sequencing, target interaction screening (ALIS/ASMS/APMS), and broad off-target counter-screening techniques (ex: CETSA/TPM))
- Experience developing and implementing native-MS methods for intact protein complex characterization.
- Experience with electron transfer dissociation (ETD) as a fragmentation technique is preferred but not required. A strong ability to evaluate the advantages and limitations of various fragmentation technologies is highly valued.
- Experience mentoring and training junior scientists.
