The Dana-Farber Cancer Institute's TPD Proteomics Core in the Fischer Lab seeks a Senior Research Technician to apply mass spectrometry-based methods to profile cellular proteomes under stimuli. You will work with technicians, scientists, postdocs, and students on projects centered on targeted protein degradation as a novel therapeutic approach.

Located in Boston, you will contribute to proteomics workflows, QC, data analysis, and protocol improvements while maintaining rigorous lab standards

Qualifications

  • Bachelor’s degree in biology, chemistry, or related field required.
  • 1–4 years in mass spectrometry or molecular biology lab (academic or industry).
  • Experience operating Thermo and/or Bruker mass spectrometers required.
  • Experience with mammalian cell culture and proteomics sample prep preferred.
  • Experience in MS data analysis and quantitative methods.
  • Knowledge of statistics and R programming preferred.
  • Familiarity with Proteome Discoverer, DIANN, and Spectronaut

Responsibilities

  • Perform cell culture and drug treatments as part of the core team.
  • Prepare proteomics samples including lysis, digestion, cleanup, and labeling.
  • Run LC-MS samples on Orbitrap/timsTOF following SOPs and QC checks.
  • Maintain lab notebooks and sample metadata accurately.
  • Collaborate with core users to ensure timely sample processing.
  • Help implement new protocols and train junior staff.
  • Maintain lab supplies and liaise with purchasing/vendors.

Job description

The Targeted Protein Degradation (TPD) Proteomics Core within the Fischer Lab at Dana-Farber Cancer Institute serves as a resource for the Cancer Biology Department, aiding investigators in answering key research questions using mass spectrometry-based proteomics.

We employ quantitative chemoproteomic and interactomic approaches to identify drug targets and are continuing to expand our repertoire to meet the needs of our core users. Our core is equipped with extensive mass spectrometry equipment including timsTOF HT, timsTOF Ultra2 and Orbitrap Eclipse tribrid mass spectrometers.

The TPD Core seeks a highly motivated individual to join the team as a Senior Research Technician, who will employ mass spectrometry-based technologies to assess the cellular proteome in response to external stimuli. This individual will be an important member of our core team and will work with technicians, scientists, postdocs and students on a variety of different research projects focused on targeted protein degradation as a novel therapeutic modality. While we consider various mass spectrometry related backgrounds, a strong interest and understanding of mass spectrometry-based proteomics is a must and strong computational skills are highly desirable.

Responsibilities:

  • Responsible for cell culture and performing drug treatments
  • Perform proteomics sample preparation including cell lysis, digestion, peptide cleanup (e.g., S-Trap, SP3), and labeling (e.g., TMT)
  • Run samples on LC-MS systems (e.g., Orbitrap, timsTOF) following standard protocols.
  • Support routine QC checks and instrument maintenance under staff supervision
  • Maintain accurate and well-organized lab notebooks and sample metadata
  • Collaborate with core users to ensure timely sample processing and communication
  • Assist in the implementation of new protocols and improvements to existing workflows
  • Help train junior staff and contribute to a clean and safe laboratory environment
  • Maintains and orders laboratory supplies
  • Communicates with purchasing and may be responsible for negotiating with vendors

Qualifications:

  • Bachelor's degree in biology, chemistry, or related field.
  • 1-4 years of experience in a mass spectrometry or molecular biology lab (academic or industry).
  • Experience operating Thermo and/or Bruker mass spectrometers is required
  • Experience with mammalian cell culture and proteomics sample preparation is preferred
  • Experience in mass spectrometry data analysis and quantitative methods
  • Knowledge of statistical and computational methods and R programming is preferred
  • Knowledge of proteomics data analysis software, including Proteome Discoverer, DIANN, spectronaut
  • Strong attention to detail and ability to follow SOPs.
