Job summary

Overview

Under the general direction of the Biotechnology Associate II, Yale Center for Genome Analysis (YCGA), the incumbent will supervise the library preparation and sequencing production sections. This role leads library preparation R&D, including protocol development and robotic implementation, in support of the Precision Medicine Initiative, and is responsible for associated instrumentation and equipment. The incumbent will establish, validate, and maintain automated workflows for library preparation, quantification, and pooling; test protocols to ensure quality and yield; and perform cell culture and nucleic acid extraction as needed.

Other Duties Include

• Coordinate daily with the Director of Operations, Research Associate II, and Research Associate III to maintain sequencer and NGS library preparation schedules and staff project assignments.
• Serve as the subject matter expert for all library preparation assays and equipment implementations.
• Establish and maintain Standard Operating Procedures (SOPs) and general policies for NGS library preparation and quality control of user-prepared libraries.
• Assist the Research Associate II with staff supervision and, as needed, serve as acting supervisor for sequencing and library preparation operations.
• Represent the Research Associate II, as needed, with the facility Administrative Supervisor on administrative matters, including staffing, temporary/casual usage, workflow and staff issue resolution, performance management, scheduling, work rules, University policy, and bargaining unit interpretation.
• Perform library and flow cell preparation/production; maintain equipment; and complete other duties as assigned.
• Troubleshoot and resolve library preparation and sequencing instrument issues, including coordination with technical support.
• Interpret results and generate reports summarizing library preparation and sequencing performance.
• Communicate with investigators and collaborators; provide technical assistance to users.
• Interact with YCGA operations teams to support efficient workflow and service delivery.
• Provide orientation and training to new lab personnel, including instruction in laboratory protocols and hands-on assistance as needed.
• Help oversee and maintain day-to-day laboratory operations.
• Conduct literature searches to stay current on sequencing and library preparation trends and proactively adapt/implement appropriate protocols.
• Assist with gathering information for grants and proposals and provide editing support.
• Support operation and maintenance of genomics instrumentation.
• Assist with ordering reagents and supplies.
• May perform other duties as assigned.

Required Skills And Abilities

• Ability to perform, assess, and troubleshoot NGS library preparation workflows (quantification, pooling, flow cell/library QC), operate and maintain sequencing/genomics instrumentation, and interpret results to generate clear reports.
• Ability to develop, implement, validate, and maintain automated/robotic library preparation protocols; optimize quality and yield; troubleshoot effectively (including working with vendor technical support); and remain proactive in improving workflows to meet deadlines.
• Ability to work independently and collaboratively; help build, lead, and support a team; provide instruction, onboarding, and guidance to a diverse group of personnel; and contribute to SOP and policy development.
• Demonstrates a high level of motivation; proficiency with basic computer programs (e.g., Word, Excel); excellent oral and written communication skills; strong interpersonal, organizational, and customer service skills; and the ability to multi-task while coordinating schedules and priorities.

Preferred Skills And Abilities

• Proven experience in molecular biology techniques such as PCR, DNA/RNA extraction and gel electrophoresis.
• Proven experience in various areas of sequencing workflow such as operation of Illumina NovaSeq instruments and NGS library preparation (single cell workflows, Olink etc).
• Proven experience with cell culture.
• Proven experience in a core lab setting with demonstrated customer service skills.
• Previous experience with NGS automation instrumentation such as Janus, Hamilton, or SciClone, Operating and maintaining genomics instruments is a plus.

Principal Responsibilities

• Actively participates in biotechnology research design, implementation, and daily operation of the lab. Develops and modifies procedures and instrument operating programs. 2. Independently carries out research projects, including experimental design, development of methods, analysis of results, and translation of results to high-throughput protocols based on results. Prepares results for discussion and reporting purposes. 3. Understands, plans, and performs complex analyses and/or syntheses of nucleic acids, proteins, or other molecules. Determines the best analytical procedures to accomplish research goals. Contributes to the development of new analysis, synthesis and technologies. 4. Programs equipment with special cycle combinations and ensures special procedures are properly executed. 5. Works closely with users during the experiment to ensure optimal outcome, including advising, responding to requests and real-time data analysis. 6. Contributes data to grant proposals and manuscripts for publication. 7. Prepares reports; responds to researchers’ more complex queries about different aspects of services provided and possible modifications. 8. May perform other duties as assigned. Required Education and Experience Master’s Degree in chemistry, biology, biochemistry, or other related discipline and one year experience or an equivalent combination of experience and education.
