Job summary

Piramal Pharma Solutions in India is seeking a Senior Research Scientist to lead reverse phase preparative HPLC purification of APIs and intermediates. The role covers method development and scale-up for challenging compounds.

You will operate and maintain preparative HPLC systems, transfer analytical methods to preparative scale, and collaborate with analytic, formulation, and regulatory teams to deliver purified compounds on schedule.

Responsibilities

• Purify APIs and related compounds using reverse phase preparative HPLC.
• Develop purification methods on C18, C8, Phenyl, Biphenyl columns.
• Scale preparative purifications from mg to gram scale.
• Design purification for poorly soluble or unstable compounds with complex impurities.
• Maximize purity, recovery and productivity within timelines.
• Transfer analytical HPLC/UPLC methods to preparative scale.
• Optimize mobile phase, pH, buffers, gradients, flow rates and load.
• Perform loading studies and scale-up purification activities.
• Improve resolution and recovery via systematic method optimization.
• Operate and maintain preparative HPLC systems and fraction collectors.
• Work with Waters, Agilent, Shimadzu, Gilson, Biotage chromatography platforms.
• Troubleshoot chromatographic systems and columns.
• Maintain instrument utilization, calibration and documentation.
• Analyze fractions using HPLC/UPLC, LC-MS and NMR.
• Interpret data to confirm identity and purity.
• Support impurity identification and structural characterization.
• Generate purification reports and summaries.
• Collaborate with chemistry, analytical, formulation, and regulatory teams.
• Prioritize multiple projects and timely delivery.
• Participate in client discussions and provide recommendations.

• Perform reverse phase preparative HPLC purification of APIs, intermediates, impurities, degradants, metabolites, and synthetic target compounds.


• Develop and optimize purification methods using C18, C8, Phenyl, Biphenyl, and specialty stationary phases.


• Execute preparative purifications ranging from milligram to multi-gram scale.


• Design and implement purification strategies for challenging compounds with poor solubility, low stability, or complex impurity profiles.


• Maximize purity, recovery, and productivity while maintaining project timelines.

Method Development and Scale-Up

• Transfer analytical HPLC/UPLC methods to preparative scale.


• Optimize mobile phase composition, pH, buffers, gradients, flow rates, and loading conditions.


• Conduct loading studies and purification scale-up activities.


• Improve chromatographic resolution and recovery through systematic method optimization.

Instrument Operation and Maintenance

• Operate and maintain preparative HPLC systems, fraction collectors, and associated software.


• Work with systems such as Waters, Agilent, Shimadzu, Gilson, Biotage, or equivalent preparative chromatography platforms.


• Perform routine troubleshooting of chromatographic systems and columns.


• Maintain instrument utilization, calibration, and documentation records.

Fraction Analysis and Characterization

• Analyze collected fractions using analytical HPLC/UPLC, LC-MS, and NMR techniques.


• Interpret chromatographic and spectroscopic data to confirm compound identity and purity.


• Support impurity identification and structural characterization activities.


• Generate purification reports and technical summaries.

Project Execution

• Support pharmaceutical R&D, process development, impurity isolation, forced degradation, and reference standard projects.


• Collaborate with synthetic chemistry, analytical, formulation, and regulatory teams.


• Prioritize multiple projects and ensure timely delivery of purified compounds.


• Participate in client discussions and provide technical recommendations.
