We are seeking a Senior Scientist in Capsid Engineering to be a key member of the Discovery Research Team. You will participate in developing Spark's gene delivery platforms, focusing on creating novel AAV capsids with enhanced properties, particularly the ability to transduce specific cell types. Discovering such capsids will facilitate the translation from in vitro to in vivo proof of concept and ultimately clinical application.
Ideate experimental strategies to engineer novel capsids with enhanced properties for clinical translation.
Develop and implement high-throughput capsid library screening strategies to discover improved variants.
Support approaches that enable the isolation of AAV capsid variants capable of transducing specific cell types in rodents and NHPs.
Collaborate with NGS and machine learning groups to enhance directed evolution for isolating AAVs with tailored properties.
Design and conduct in vitro studies in preclinical animal models of genetic diseases in collaboration with other Discovery and Gene Therapy scientists.
Support joint programs and collaborations with external partners.
Write study reports, manuscripts, and present results at scientific meetings.
PhD in a biological discipline with 3-5 years of relevant experience.
Hands-on experience with high complexity library preparations.
Familiarity with NGS and bioinformatics is required.
Proficiency in lab techniques such as plasmid design, molecular cloning, qPCR, cell culture, ELISA, luciferase assays, SDS-PAGE/Western blotting, etc.
Experience with AAV gene therapy is required.
Willingness to travel occasionally for short periods.
Experience with laboratory animal studies is preferred.
Strong analytical and problem-solving skills.
Familiarity with bio/pharma software tools.
Strong publication record is a plus.
Experience managing external collaborations is a plus.
The expected salary range for this position in Philadelphia is $103,400 to $192,000. Actual pay will be determined based on experience, qualifications, location, and other factors permitted by law.
