CRISPR Therapeutics, Inc. in Boston is seeking a Senior Oligonucleotide Scientist to lead design, synthesis, purification, and analysis of complex, highly modified oligonucleotides.

You will drive scale-up, purification optimization, and analytical characterization to support pipeline advancement. The role emphasizes solid-phase synthesis expertise, state-of-the-art purification, and mentoring junior scientists within a on-site team.

Qualifications

• Ph.D. in Organic Chemistry or Medicinal Chemistry with 4.5–8+ years of industry biopharma/biotech experience or non-PhD with 12–15 years of progressive experience.
• Deep understanding of phosphoramidite chemistry, protecting groups, and nucleotide modifications (e.g., 2'-OMe, 2'-F, phosphorothioates, LNA).
• Hands-on experience with automated oligonucleotide synthesizers and preparative purification systems.

Responsibilities

• Lead automated solid-phase synthesis of highly modified oligonucleotides at various scales.
• Develop purification processes using RP-HPLC, IEX and SEC, plus TFF and lyophilization.
• Oversee analytical characterization using LC-MS, MALDI-TOF, NMR, and capillary electrophoresis.
• Drive optimization of synthetic routes, yields, and scalable, reproducible processes.
• Evaluate novel chemical modifications and conjugation strategies to enhance stability and potency.
• Serve as oligochemistry lead on multidisciplinary project teams.
• Mentor junior chemists and document findings for regulatory filings.
