Job summary

Flagship Pioneering, Cambridge-based FL130, is hiring a Principal Scientist to design and run high-throughput pooled screening campaigns. You will own library design, FACS-based selection, sequencing readouts, and data analysis from day one, shaping the company’s scientific direction.

The role offers ownership of the technical direction, opportunities to build a team, and collaboration with translational colleagues across Flagship portfolio companies.

Qualifications

• PhD in molecular or cell biology, genetics, bioengineering, or a related field with extensive postdoc/industry experience.
• Hands-on experience running high-throughput pooled screens with FACS-based selection.
• Experience designing DNA and oligo libraries for high-throughput screening.
• Strong flow cytometry and cell-sorting expertise.
• Experience with lentiviral transduction and sequencing workflows.

Responsibilities

• Plan, execute, and analyze high-throughput pooled screens using FACS as the primary selection modality.
• Design and build custom DNA and oligo libraries for high-throughput pooled screening.
• Develop and optimize FACS-based selection strategies across various cell lines.
• Culture and screen in primary and primary-like cell models, including difficult-to-transduce lines.
• Produce and titer lentiviral libraries; optimize MOI and coverage.
• Prepare and process sorted populations for sequencing and barcode-readout workflows.
• Analyze sequencing data to call hits and assess enrichment.
• Optimize workflows and maintain rigorous documentation practices.
• Collaborate with protein-engineering and translational teams to align library design to validation.
• Translate data into insights and present at team meetings.
