Lilly's RNA Therapeutics organization in South San Francisco seeks a computational biophysicist to build atomic-resolution models of Ago2 guide-strand loading and catalytic activity, translating data into design rules for medicinal chemists.

You will run MD and free-energy simulations, integrate cryo-EM, crystallography and biophysical data, and collaborate with chemistry, structural biology and biochemistry teams to shape oligonucleotide modifications across programs.

Formación

• PhD in biophysics, computational chemistry, chemical physics, structural biology or a related STEM field.
• Five+ years running and analyzing MD simulations of protein–nucleic acid systems.
• Experience with MD engines and free-energy methods (FEP, TI, umbrella sampling, metadynamics).

Responsabilidades

• Model how chemical modifications affect Ago2 binding and RISC catalytic efficiency.
• Run all-atom and enhanced-sampling simulations of siRNA complexes and RISC intermediates.
• Quantify loading energetics and conformational dynamics of guide strands.
• Model seed‑mediated off-target engagement and modification balance.
• Convert predictions into design hypotheses and guide medicinal chemistry.
• Validate predictions against ITC, SPR, DSC and functional data.
• Build simulation pipelines and force-field parameters for non-natural nucleotides.
• Present findings and recommendations to chemistry, biology and leadership.
