BCforward seeks a Scientist II to advance high-throughput LVV and AAV vector production with automation integration and rapid screening for pipeline studies. The role covers upstream/downstream development, QC analytics, and data-driven optimization in a fast-paced setting.
Ideal candidates have experience in transfection, DOE, and ex vivo CAR T engineering using lentivirus, with strong data presentation and cross-functional collaboration skills. On-site in South San Francisco, CA.
We are seeking a Scientist II to join our dynamic team. The ideal candidate will have strong experience in high-throughput LVV and AAV viral vector production, upstream and downstream process development, and QC analytics and a proven ability to increase vector production throughput and yield, integrate automation, and deliver rapid turnaround for screening and pipeline studies.
Responsibilities:
Integrate automation into high-throughput LVV and AAV workflows. Program automated liquid handlers (e.g., Integra Assist Plus, Bravo) for transfection, transduction, dilution, and cell seeding. Incorporate shaker arrays or micro-scale bioreactors (e.g., Ambr 15, Ambr 250) for parallel mini-scale runs. Troubleshoot liquid classes, pipetting precision, and plasticware compatibility to minimize variability.
Develop and optimize a suspension-based LVV upstream platform from seed train through shake flask production and primary harvest. Apply DOE to evaluate multivariate factors including transfection conditions, plasmid ratios, reagents (e.g., PEI, lipid nanoparticles), DNA-to-cell ratios, harvest timing, feed schedules, and chemical enhancers.
Establish downstream LVV processes. Evaluate low-shear depth filtration and microfiltration protocols. Develop and optimize TFF and chromatographic purification methods.
Conduct QC analytics for LVV and AAV. Perform physical and functional characterization to define quality attributes. Integrate QC into upstream and downstream development for rapid feedback and improvement.
Manage data, inventory, and protocols. Analyze multivariate datasets using Excel, Prism, or AI tools. Maintain a structured virus inventory and registry in digital platforms. Author SOPs, technical reports, and tech transfer documents. Present data in group and department meetings.
Required Skills & Qualifications:
Proficiency in mammalian cell culture and molecular biology, including transient plasmid transfection, PCR, qPCR, ddPCR/dPCR, and Western blot.
Strong data analysis and presentation skills with the ability to interpret multivariate process datasets.
Hands-on experience in LVV or AAV production in adherent or suspension cells and process development.
Experience with ex vivo CAR T engineering using lentivirus.
Preferred Skills:
Experience with statistical DOE modeling and automated liquid handling platforms.
Familiarity with flow cytometer operations, electronic lab notebooks, and BSL2 facility practices.
