We offer an exciting opportunity to conduct your Master Thesis on the following topic:

Development of a robust reducing peptide mapping workflow to characterize mAb via multi-attribute method (MAM)

The multi-attribute-method (MAM) is a liquid chromatography-mass spectrometry (LC-MS) based method used to comprehensively characterize critical quality attributes (CQAs) at the amino acid level of biopharmaceuticals. The MAM is based on the reduced peptide mapping (rPepmap) workflow, which involves disulfide linkage reduction, alkylation, proteolytic digestion, LC separation, and MS detection and identification. To develop a reliable MAM, an optimized rPepmap workflow must be implemented to enhance robustness and reproducibility of LC-MS data for identifying and quantifying CQAs.

This research focuses on:

  • Optimizing sample preparation for protein digestion to reduce methionine-related artifacts, decrease column regeneration efforts, and improve protein sequence coverage. Testing SP3 magnetic beads-based methods for protein digestion and buffer exchange.
  • Optimizing MS and MS/MS acquisition parameters to improve peptide fragmentation, identification, and the quantification of post-translational modifications (PTMs).
  • Conducting a feasibility study to assess the robustness and reproducibility of the workflow following ICH Q2(R2) guidelines, including sensitivity, accuracy, specificity, and robustness.
  • Applying the established LC-MS workflow to characterize model proteins (mAbs and variants or impurities).

The workflow will be based on existing rPepmap methods using 1-2 mAbs as models. Samples will be analyzed on a Vanquish UPLC or nanoLC coupled to a Fusion Orbitrap MS system capable of HCD and ETD.

Eligibility Requirements

  • Postgraduate in analytical chemistry, chemical biology, biochemistry, or related fields.
  • Basic knowledge of mass spectrometry; practical experience is advantageous.
  • Excellent command of English (spoken and written).
  • Flexible, committed, and self-organized working style.
