The research plan for this post-doctoral position funded by the São Paulo Research Foundation (FAPESP) focuses on studying GRASP proteins, particularly their structural and dynamic properties. It involves a multi-faceted approach using several experimental methods, such as X-ray crystallography and Cryo-electron microscopy (Cryo-EM) to analyze GRASPs from different organisms (including human GRASPs), especially under conditions associated with unconventional protein secretion (UPS). GRASP proteins are essential for Types III and IV UPS in eukaryotic cells, and the structure of the Golgi apparatus. The project aims to provide insights into GRASP's structural organization, particularly the underexplored C-terminal SPR domain, which poses challenges due to its disordered nature.
Key objectives include obtaining high-resolution structural data, examining GRASP protein structure and dynamics, and exploring in vitro and in-cell structural behavior. Collaborations with experts in crystallography, Cryo-EM, and cell biology will support these goals. The project promotes interdisciplinary training in molecular biophysics, biochemistry, and cell biology, aligning with the FAPESP PD program's objectives to provide early-career researchers with a broad range of scientific expertise. Training in heterologous expression, protein purification, and X-ray crystallography or cryo-EM is required.

This opportunity is open to candidates of any nationality. The selected candidate will receive a FAPESP Post-Doctoral fellowship in the amount of R$ 12,000.00 monthly and a research contingency fund, equivalent to 10% of the annual value of the fellowship which should be spent on items directly related to the research activity.
