A post-doctoral position is open in the Laboratory of Biomolecular NMR Spectroscopy at CEPID B3 (Biology of Bacteria and Bacteriophages Research Center), a Research, Innovation and Dissemination Center (“CEPID” in the Portuguese language acronym) supported by the São Paulo Research Foundation (FAPESP) and hosted by the University of São Paulo's Institute of Chemistry (IQ-USP) in Brazil. The project focuses on Type IV Secretion Systems (T4SSs), which include conjugation systems (DNA transfer between bacteria) and effector translocators (protein delivery into cells).
T4SSs comprise 11 conserved VirB subunits and VirD4, organized in the outer membrane core complex (OMCC) and inner membrane complex (IMC). The T4SS of Xanthomonas citri (T4SS XAC) is specialized in secreting toxins that kill gram-negative bacteria, functioning as a bacterial weapon. VirD4, a membrane-associated hexameric ATPase, recruits substrates for secretion. All T4SS XAC substrates (X-Tfes) contain a flexible ~120-aa C-terminal domain (XVIPCD) that binds tightly to the VirD4 All-Alpha-Domain (AAD). To elucidate recruitment and translocation mechanisms, we will analyze the structure and dynamics of X-Tfes and their interaction with VirD4 using nuclear magnetic resonance (NMR), X-ray crystallography, and ITC.
Candidates must hold a PhD in biochemistry, molecular biophysics, or related areas, with experience in protein NMR and crystallography. Skills in cloning, expression, and purification are a plus.
