Job summary

Stoke Therapeutics is seeking a Senior Research Associate in In Vivo Pharmacology to join our preclinical team in Bedford, MA (moving to Waltham, MA). You will execute complex rodent studies using antisense oligonucleotides to restore protein expression, perform neonatal and adult injections, and support PK/PD and efficacy analyses with rigorous data handling.

Required experience includes rodent dose administration routes (SC/IV/IT/ICV), necropsy, anesthesia, and surgical skills.

Qualifications

• BS in Pharmacology or Neuroscience with 4 years experience, MS with 2 years preferred.
• Experience with rodent dose administration routes (SC, IV, IT, ICV).
• Experience with PK/PD studies in rodents.
• Strong necropsy and anesthesia skills.
• Surgical skills including stereotaxic ICV injections.
• Experience with rodent behavioral assays (Open Field, DigiGait, Rotarod).
• Familiarity with Softmouse colony management software.
• Proficiency in data analysis tools (Excel, GraphPad, PowerPoint).

Responsibilities

• Design and execute in vivo rodent studies to advance preclinical pipeline using antisense oligonucleotides to restore protein expression.
• Perform rodent procedures including bolus ICV and IT injections in neonate and adult mice and rats; other routes include SC and IV with blood collection, serial sampling, and tissue collections.
• Perform adult surgical procedures including dosing, anesthesia monitoring and pre/post-op preparations.
• Execute dose-response, time-course, PK/PD, tolerability and efficacy studies.
• Maintain breeding colonies, wean and genotype, and utilize Softmouse to expand colonies for experiments.
• Assess animal phenotypes by implementing behavioral assays like Open Field, DigiGait, Rotarod.
• Prepare tubes for sample collections and perform necropsies and tissue homogenization and molecular biology assays.
• Help analyze and interpret experimental data using appropriate statistical approaches and software.
• Maintain accurate laboratory records, electronic notebooks, study protocols, and data reports.
• Other tasks as assigned

About Stoke:

Position Purpose:

Stoke Therapeutics is seeking a highly skilled and enthusiastic Senior Research Associate in In Vivo Pharmacology to join our preclinical research team. This individual will be instrumental in the execution of in vivo studies in a fast-paced and exciting environment working with rodent models in the CNS and other therapeutic programs. This position will report to the Associate Director of In Vivo Strategy and Operations.

Key Responsibilities:

• Work cross-functionally with scientists to help design and execute complex in vivo rodent studies to advance Stoke’s preclinical pipeline using antisense oligonucleotides to restore protein expression.
• Perform hands-on rodent procedures including bolus ICV and IT injections in both neonate and adult mice and rats. Other routes of injections include subcutaneous and intravenous injections with subsequent blood collection, serial sampling, and tissue collections.
• Perform adult surgical procedures including dosing, anesthesia monitoring and pre/post-op preparations.
• Execute dose-response, time-course, PK/PD, tolerability and efficacy studies.
• Maintain breeding colonies, wean and genotype, and utilize Softmouse to help expand colonies for experiments.
• Assess animal phenotypes by implementing various behavioral assays like Open Field, DigiGait, and Rotarod.
• With a keen eye for attention to detail, prepare tubes for sample collections and perform necropsies and subsequent tissue homogenization and molecular biology assays.
• Help analyze and interpret experimental data using appropriate statistical approaches and software.
• Maintain accurate laboratory records, electronic notebooks, study protocols, and data reports.
• Other tasks as assigned

Required Skills & Experience:

• S. with 4 years of experience or M.S. with 2 years of experience (relevant industry experience is a plus). Degrees in Pharmacology, Neuroscience or relevant disciplines highly preferred.
• Proficient in various routes of dose administration in mice and rats including, but not limited to, SC, IV, IT and ICV.
• Experience with PK/PD studies in rodents
• Serial sampling in rodents is a plus
• Extensive knowledge of rodent anatomy to perform large scale necropsies
• Experience with anesthesia and pre- and post-op monitoring
• Skilled in surgical techniques such as stereotaxic ICV injections
• Knowledge with rodent behavioral studies of the CNS
• Experience with breeding and colony management
• Tissue processing and homogenization
• qPCR, ELISA, MSD
• Familiarity with Softmouse colony management Software is a plus
• Proficient with Excel, GraphPad Prism, and PowerPoint for data interpretation and presentation
• Electronic Notebook
• Familiarity with writing technical reports
• Strong aptitude for adapting technological advances in the field

Location(s):

Stoke is located in Bedford, MA, and will be moving to a new location in Waltham, MA by the end of 2026. This position is a lab-based position currently in the Bedford office and will later be based in Waltham.

Travel:

This position will require approximately 5% travel.

The anticipated salary range for this role is $98,000 - $110,000. The final offer will be determined based on a variety of factors, including role scope, experience and qualifications, education, market benchmarks, internal equity, and geographic location.
